Overview

delta-[1-13C] gluconolactone is a carbon-13 isotopically labeled research imaging probe used in hyperpolarized magnetic resonance spectroscopy (MRS) to assess metabolic flux through the pentose phosphate pathway (PPP). The compound is a labeled version of delta-gluconolactone (D-glucono-1,5-lactone) that enters cells via glucose transporters and is subsequently phosphorylated to 6-phospho-delta-[1-13C]gluconolactone. It serves as a substrate for the enzyme 6-phosphogluconolactonase (PGLS), which converts it to [1-13C]6-phosphogluconate (6PG). This metabolic conversion allows researchers to visualize dynamic PPP activity in vivo. Preclinical studies, primarily conducted at institutions like UCSF, have utilized this probe to image glioblastoma (GBM), specifically to monitor TERT-associated changes in redox metabolism and evaluate response to therapies such as temozolomide or GABPB1 targeting.
